浊度对荧光光谱法测定水体中叶绿素a的影响

摘要: 研究了浊度对荧光光谱法测定水体中叶绿素a的影响.采用荧光分光光度计分别测定了不同浊度的叶绿素a提取液和铜绿微囊藻液的荧光强度,使用海水叶绿素现场检测仪测定了不同浊度的铜绿微囊藻液和实际水样的叶绿素a浓度值.结果表明:当水体浊度较大时,由于悬浮颗粒物的强烈散射作用会使叶绿素a的测定结果偏高.当水体透明度较高时,浊度对测定的影响基本可以忽略.Abstract: Effect of turbidity on the fluorescence intensity determination of chlorophyll-a in water was studied. Spectrofluorophotometer was used to measure the the fluorescence intensity of the solution microcystis aeruginosa and the extract of chlorophylla with different turbidity. The concentration of chlorophyll-a in microcystis aeruginosa and water samples with different turbidity was determined using the on-line detector for chlorophyll in seawater. It was found that the measured concentration of chlorophyll-a was high due to the strong scattering effect of suspended particules when water turbidity was high. The effect of turbidity was negligble when the water was transparent.
