    氨基甲磺酸席夫碱-邻菲啰啉镍(Ⅱ)配合物与DNA反应的共振散射光谱的研究及应用

    Study and Application of Resonance Light Scattering Spectroscopy Produced by the Reaction of Aminomethylsulfonic Acid Schiff Base-Phen-Ni(Ⅱ) Complex with Deoxyribonucleic Acid

    • 摘要: 氨基甲磺酸席夫碱-邻菲啰啉镍(Ⅱ)配合物与脱氧核糖核酸相互作用的共振散射光谱在311 nm处体系有共振光散射,据此提出了一种测定脱氧核糖核酸的新方法.优化的试验条件如下:① 缓冲溶液的pH为7.5;② 反应温度为37.5 ℃;③ 反应时间为25 min;④ 配合物的用量为1.1 mL.脱氧核糖核酸的线性范围为0.53~7.87 mg·L-1,方法的检出限为0.16 mg·L-1.方法用于人工试样的分析,回收率在99.7%~102%之间,测定值的相对标准偏差(n=9)为1.7%.

       

      Abstract: The resonance light-scattering spectrum of the reaction of aminomethylsulfonic acid schiff base-phen-Ni(Ⅱ) complex and deoxyribonucleic acid has a resonance peak at 311 nm. Based on this fact,a new method for determination of deoxyribonucleic acid was proposed. The optimized conditions were as follows: ① pH of buffer solution: 7.5;② reaction temperature: 37.5 ℃;③ reaction time: 25 min;④ amount of the complex: 1.1 mL. The linear range of deoxyribonucleic acid was 0.53-7.87 mg·L-1 with detection limit of 0.16 mg·L-1. The proposed method was applied to the analysis of synthetic samples,giving values of recovery in the range of 99.7%-102% and RSD (n=9) of 1.7%.
