氯化血红素催化-荧光光谱法测定染发剂中的过氧化氢
Determination of Hydrogen Peroxide in Hair Dye by Fluorescence Spectrometry Base on Hemin Catalysis

摘要: 利用过氧化氢能够在氯化血红素作为模拟酶催化条件下将对羟基苯乙酸氧化成具有较强荧光的二聚体的特点,建立了荧光光谱法测定染发剂中过氧化氢的方法。氯化血红素的浓度为10 μmol·L-1,对羟基苯乙酸浓度为1 mmol·L-1,pH 10,反应温度为室温,反应时间5 min,激发波长为310 nmm,发射波长为400 nm,体系的荧光强度达到最大。过氧化氢的浓度在0.05~50 μmol·L-1范围内与体系的荧光强度呈线性关系,检出限(3S/N)为0.3 μg·kg-1。加标回收率在94.5%~105%之间,测定值的相对标准偏差(n=6)小于2%。Abstract: A method for the determination of hydrogen peroxide in hair dye by fluorescence spectrometry was developed based on that, the p-hydroxyphenylacetic acid could be oxidized by hydrogen peroxide with hemin as mimetic catalyzer to form the dimer with strong fluorescence. The optimum conditions for maximum fluorescence intensity were as follow: 10 μmol·L-1 of hemin, 1 mmol·L-1 of p-hydroxyphenylacetic acid, pH 10, reaction at room temperature for 5 min, λex of 310 nm and λem of 400 nm. Linear relationship was found between the fluorescence intensity and the concentration of hydrogen peroxide in the range of 0.05-50 μmol·L-1, with detection limit (3S/N) of 0.3 μg·kg-1. Recovery rates obtained by standard addition method were in the range of 94.5%-105% with RSDs (n=6) less than 2%.
