荧光光谱法测定氧氟沙星的含量
Determination of Ofloxacin by Fluorescence Spectrometry

摘要: 室温下,4.0×10-6mol·L-1氧氟沙星溶液5 mL与8.0×10-6mol·L-1茜素红溶液5 mL发生电荷转移反应30 min后生成稳定的络合物,并能产生荧光,络合物的最大激发波长为303 nm,最大发射波长为508 nm,络合物的组成为1比1。基于此建立了测定氧氟沙星的荧光光谱法。氧氟沙星的浓度在2.0×10-7~4.0×10-6mol·L-1范围内与络合物的荧光强度呈线性关系,方法的检出限为9.1×10-9mol·L-1。加标回收率在96.0%~101%之间,测定值的相对标准偏差(n=5)小于2%。利用本法测定氧氟沙星药片中氧氟沙星的含量,测定值与标示值相符。Abstract: At room temperature, 5 mL of ofloxacin solution (4.0×10-6mol·L-1) reacted with 5 mL of alizarin red solution (8.0×10-6mol·L-1) for 30 min to form a stable charge-transfer complex, which showed fluorescence. The maximum excitation wavelength of the complex was 303 nm, and the maximum emission wavelength was 508 nm. The composite ratio of ofloxacin and alizarin red in the complex was 1∶1. Based on that, a method of fluorescence spectrometry was established for the determination of ofloxacin. A linear relationship was found between the fluorescence intensity and the concentration of ofloxacin in range of 2.0×10-7-4.0×10-6mol·L-1, with detection limit of 9.1×10-9mol·L-1. Recovery rates obtained by standard addition method were in the range of 96.0%-101% and RSDs (n=5) was less than 2%. The method was applied to the determination of ofloxacin in pharmaceutical formulation, giving results in consistent with the marked values.
