超高效液相色谱-串联四极杆质谱法测定鸡腿菇中4-己基间苯二酚的含量
UPLC-MS/MS Determination of 4-Hexylresorcinol in Coprinus Comatus

摘要: 采用超高效液相色谱-串联四极杆质谱法测定鸡腿菇中4-己基间苯二酚的含量。试样经乙腈超声萃取,提取液经SPE-C18柱净化后,在ACQUITY UPLC BEH C18色谱柱上分离,以乙腈-0.1%(φ)甲酸溶液为流动相进行梯度洗脱,质谱中选择电子轰击离子源和多反应监测模式。4-己基间苯二酚的质量浓度在5.0~500.0 mg·L-1范围内与色谱峰面积呈线性关系,检出限(10S/N)为1.0 μg·kg-1。加标回收率在86.7%~94.4%之间,测定值的相对标准偏差(n=6)小于2.2%。Abstract: UPLC-MS/MS was applied to the determination of the residues of 4-hexylresorcinol in fresh Coprinus Comatu. The sample was ultrasonically extracted with acetonitrile, and the extract solution was purified with SPE-C18 column. The purified solution was separated on an ACQUITY UPLC BEH C18 column, with a mixture of acetonitrile and 0.1% (φ) formic acid solution as mobile phase for gradient elution. EI and MRM were adopted in MS. Linear relationship was found between peak areas and the mass concentration of 4-hexylresorcinol in the range of 5.0-500.0 mg·L-1, with detection limit (10S/N) of 1.0 μg·kg-1. Recovery rates obtained by standard addition method were in the range of 86.7%-94.4%, and RSDs (n=6) were less than 2.2%.
