    手性流动相添加剂-超高效液相色谱法拆分头孢噻吩对映体

    UHPLC Separation of Cephalothin Enantiomers with Chiral Mobile Phase Additives

    • 摘要: 在流动相中加入手性选择剂羟丙基-β-环糊精(HP-β-CD)实现了用超高效液相色谱法拆分头孢噻吩对映体。所用色谱柱为Agilent Poroshell 120 SB-C18色谱柱(2.1 mm×100 mm,2.7 μm);流动相为乙腈与pH 7.2的12.0 mmol·L-1羟丙基-β-环糊精溶液以体积比80:20组成的混合液;流量为0.25 mL·min-1。在所选条件下头孢噻吩的对映体获得基线分离,连续测定6次的相对标准偏差不大于2.3%。

       

      Abstract: Hydroxypropyl-β-cyclodextrin (HP-β-CD) was chosen as a chiral selector to be added to the mobile phase to achieve separation of enantiomers of cephalothin by UHPLC. Agilent Poroshell 120 SB-C18 (2.1 mm×100 mm, 2.7 μm) chromatographic column was used as stationary phase. A mixture of acetonitrile and 12.0 mmol·L-1 HP-β-CD solution (pH 7.2), mixed in the ratio of 80 to 20, was used as mobile phase with a flow rate of 0.25 mL·min-1. Under the chosen conditions, good baseline separation of the enantiomers of cephalothin was attained, and giving values of RSD's obtained from 6 successive determinations no more than 2.3%.
