    菊花的气相色谱指纹图谱及其系统聚类分析

    Study on the Gas Chromatographic Fingerprints and Hierarchical Cluster Analysis for Chrysanthemum

    • 摘要: 收集了4个产地的12个菊花样品,用顶空-固相微萃取-气相色谱法(HS-SPME-GC)在选定的条件(萃取温度80 ℃,萃取时间30 min,选择65 μm PDMS/DVB作为萃取头)测得其挥发组分的色谱图,在所得到的50个色谱峰中,选择了22个共有峰,并选择其中的9号峰(保留时间25.57 min)为参照峰。分别求得各峰的α值和S值后,按规定的技术要求建立了12个菊花样品的指纹图谱。经质谱分析对比NIST 08谱库并结合相关文献对主要挥发性成分作了鉴定。通过SPSS 16.0软件用系统聚类分析对上述指纹图谱进行分类比较,应用所提出的方法对2个市售菊花样品作出产地鉴别和质量评价。

       

      Abstract: Twelve samples of chrysanthemum grown in 4 different areas in China were collected and sampled as described, and analyzed by HS-SPME-GC under the optimum conditions (temperature and time of extraction: 80 ℃ and 30 min; 65 μm PDMS/DVB microextractor was selected for extraction) to obtain the chromatogram of their volatile components. Among the 50 peaks found, 22 common peaks were selected and the peak (No. 9) with retention time of 25.57 min was selected as the reference peak. Values of α and S were calculated on the base of peak areas according to the technical specification, and the fingerprints of the 12 chrysanthemum samples were established. Mass spectroscopy was applied to characterize the volatile components in the chrysanthemum samples by referring to the NIST 08 spectrographic data base and related literatures. Furthermore, hierachical cluster analysis was applied with the aid of the software of SPSS 16.0 to have the fingerprints studied comparatively and classified. By applying the proposed method to analyze 2 unknown chrysanthemum samples, growing areas of the samples were identified and their quality were evaluated.
