    金属有机框架-有序介孔碳修饰的玻碳电极用于测定8-羟基脱氧鸟苷

    Determination of 8-Hydroxy-2'-deoxyguanosine Using Glassy Carbon Electrode Modified with Metal Organic Framework and Ordered Mesoporous Carbon

    • 摘要: 通过水热法制备了金属有机框架材料(ZIF-8),采用ZIF-8和有序介孔碳(OMC)修饰玻碳电极(GCE),采用循环伏安法和交流阻抗法对此修饰电极(ZIF-8/OMC/GCE)的电化学性能进行了研究。结果表明,经过修饰可增大裸玻碳电极的有效表面积、改善电极的电催化活性。利用差分脉冲伏安法研究了8-羟基脱氧鸟苷在ZIF-8/OMC/GCE上的电化学特性。结果表明,此修饰电极的电流响应值与8-羟基脱氧鸟苷的浓度在0.35~350 μmol·L-1内呈线性关系,检出限(3S/N)为0.22 μmol·L-1。采用修饰电极测定人尿样中的8-羟基脱氧鸟苷,加标回收率在90.1%~108%之间。

       

      Abstract: A metal organic framework material (ZIF-8) was prepared by a hydrothermal method. The glassy carbon electrode (GCE) was modified by ZIF-8 and ordered mesoporous carbon (OMC). Electrochemical behavior of the modified electrode (ZIF-8/OMC/GCE) was studied by cyclic voltammetry and alternating current impedance, and as shown by the results, the effective surface area of the modified electrode was increased significantly as compared with the bare GCE, thus leading to improvement of electrocatalytic activity of the modified electrode. Electrochemical characteristics of 8-hydroxy-2'-deoxyguanosine (8-OHdG) at ZIF-8/OMC/GCE were studied by differential pulse voltammetry, and it was found that linear relationship was obtained between values of current response and concentration of 8-OHdG in the range of 0.35-350 μmol·L-1, with the detection limit (3S/N) of 0.22 μmol·L-1. The modified electrode was used to determine 8-OHdG in human urine samples, giving values of recovery obtained by standard addition method in the range of 90.1%-108%.
