    微波密闭消解-电感耦合等离子体质谱法测定小鼠血浆中硒的含量

    Determination of Selenium in Mouse Plasma by Microwave Sealed Digestion Coupled with Inductive Coupled Plasma Mass Spectrometry

    • 摘要: 移取小鼠血浆样品100 μL于消解罐中,加入5 mL硝酸与2 mL 30%(质量分数)过氧化氢溶液进行微波密闭消解,冷却后,将样品溶液赶酸至少于0.5 mL,用水定容至25 mL,以73Ge为内标,选用标准检测模式(STD)。硒的线性范围为0.2~20 μg·L-1,检出限(3s)为6.75 μg·L-1。加标回收率在93.1%~105%之间,测定值的相对标准偏差(n=6)小于5.0%。利用本方法测定补硒小鼠血浆中的硒含量,可观察到硒含量随给药时间而变化。

       

      Abstract: 100 μL of mouse plasma sample was added into the digestion tank, and 5 mL of nitric acid and 2 mL of 30% (w) hydrogen peroxide solution were added for microwave sealed digestion. After cooling, the sample solution was treated to volatilize acid to less than 0.5 mL, and then diluted to 25 mL with water. 73Ge was used as the internal standard and the standard detection mode (STD) was selected. The linear range of selenium was 0.2-20 μg·L-1, and the detection limit (3s) was 6.75 μg·L-1. Values of recovery obtained by standard addition method were between 93.1% and 105%, and the relative standard deviation (n=6) of the measured values was less than 5.0%. The method was used to determine the selenium content in the plasma of selenium supplemented mice, and results showed that the selenium content changed with the administration time.
