    共振光散射光谱法测定牛血清蛋白

    Determination of Bovine Serum Albumin by Resonance Light Scattering Spectroscopy

    • 摘要: 于10 mL比色管中依次加入pH 3.29的Britton-Robinson缓冲溶液3.00 mL,一定体积的8.8×10-6mol·L-1牛血清蛋白溶液,1.00×10-3mol·L-1的2-2-(5,6-二氯苯并噻唑)偶氮-5-二甲氨基苯甲酸溶液0.40 mL,再加入35 g NaCl,以水定容,摇匀,在35℃反应3 h后,在激发波长和发射波长均为524 nm处测定体系的共振光散射强度。结果发现,牛血清蛋白的浓度在1.00~25.00 μmol·L-1内与体系的共振光散射强度呈线性关系,检出限(3S/N)为0.33 μmol·L-1。方法用于测定牛尿中牛血清蛋白的含量,加标回收率在99.6%~102%之间,测定值的相对标准偏差(n=6)在1.8%~5.8%之间。

       

      Abstract: 3.00 mL of Britton-Robinson buffer solution (pH 3.29), certern volume of 8.8×10-6mol·L-1 bovine serum albumin solution, and 0.40 mL of 1.00×10-3mol·L-1 2-2-(5,6-dichlorobenzothiazolyl)azo-5-dimethylaminobenzoic acid solution, were added into a 10 mL colorimetric tube successively. After 35 g of NaCl was added, the mixture was made up to 10 mL with water, shaked well, and reacted at 35℃ for 3 h. Then the resonance light scattering intensity of the system was mesured at the excitation wavelength and the emission wavelength of 524 nm. Linear relationship was found between the resonance light scattering intensity of the system with the concentration of bovine serum albumin in the range of 1.00-25.00 μmol·L-1, and the detection limit (3S/N) was 0.33 μmol·L-1. The method was applied to determination of the content of bovine serum albumin in bovine urine, gving values of recovery between 99.6% and 102%, and RSDs (n=6) between 1.8%-5.8%.
