    超高效液相色谱-串联质谱法测定饲料中氨苯胂酸和洛克沙胂

    UHPLC-MS/MS Determination of Arsanilic Acid and Roxarsone in Feed

    • 摘要: 采用超高效液相色谱-串联质谱法测定饲料中氨苯胂酸和洛克沙胂的含量。粉碎过筛后的样品用乙腈(1+1)溶液超声提取30 min,离心后,上清液用二氯甲烷净化,取水层(上层)过0.22 μm水系滤膜。以Thermo HYPERCARB色谱柱为固定相,以不同体积比的0.05%(体积分数)甲酸溶液和乙腈混合液为流动相进行梯度洗脱,串联质谱分析中采用电喷雾负离子源和多反应监测模式。氨苯胂酸和洛克沙胂的质量浓度均在0.05~10.0 mg·L-1内与其对应的峰面积呈线性关系,检出限(3S/N)为0.01~0.05 mg·kg-1。以空白样品为基体进行加标回收试验,所得回收率为82.0%~106%,测定值的相对标准偏差(n=6)为1.0%~7.9%。

       

      Abstract: UHPLC-MS/MS was applied to the determination of arsanilic acid and roxarsone in feed. The crushed and screened sample was extracted ultrasonically for 30 min with acetonitrile (1+1) solution. After centrifugation, the supernatant was purified with methylene chloride. The water layer (upper level) was filtered on 0.22 μm filtering membrane of water system. Thermo HYPERCARB chromatographic column was used as stationary phase, and the mixture of 0.05% (φ) formic acid solution and acetonitrile mixed in different ratios was used as mobile phase in gradient elution. ESI- and multi-reactions monitor mode were adopted in MS/MS. Linear relationships between values of peak area and mass concentration of arsanilic acid and roxarsone were kept in the same range of 0.05-10.0 mg·L-1, with detection limits (3S/N) in the range of 0.01-0.05 mg·kg-1. On the base of blank sample, test for recovery was made by standard addition method; values of recovery found were in the range of 82.0%-106%, with RSDs (n=6) of determination values in the range of 1.0%-7.9%.
