    亲水作用色谱法同时测定食品添加剂中D-异抗坏血酸、L-抗坏血酸和二氧化硫脲

    Simultaneous Determination of D-iso-Ascorbic Acid, L-Ascorbic Acid and Thiourea Dioxide in Food Additives by Hydrophilic Interaction Chromatography

    • 摘要: 食品添加剂样品用含1%(体积分数,下同)甲酸的100 mmol·L-1甲酸铵溶液溶解,经乙腈稀释后,用0.22 μm微孔滤膜过滤,采用亲水作用色谱法同时测定滤液中D-异抗坏血酸、L-抗坏血酸和二氧化硫脲的含量。以InfinityLab Poroshell 120 HILIC-Z色谱柱(3.0 mm×100 mm,2.7 μm)为固定相,以含1%甲酸的100 mmol·L-1甲酸铵溶液和乙腈以体积比15∶85组成的混合液为流动相,用二极管阵列检测器测定。D-异抗坏血酸、L-抗坏血酸和二氧化硫脲的质量浓度均在1.0~100 mg·L-1内与其对应的峰面积呈线性关系,检出限(3S/N)均为1.0%。以空白样品为基体进行加标回收试验,所得回收率为94.7%~108%,测定值的相对标准偏差(n=6)小于8.0%。

       

      Abstract: The food additive sample was dissolved with 100 mmol·L-1 ammonium formate solutioncontaining 1% (φ, the same below) formic acid. After dilution with acetonitrile, the sample solution was filtered with 0.22 μm millipore membrane. Hydrophilic interaction chromatography was applied to the simultaneous determination of D-iso-ascorbic acid, L-ascorbic acid and thiourea dioxide in the filtrate. InfinityLab Poroshell 120 HILIC-Z chromatographic column (3.0 mm×100 mm, 2.7μm) was used as stationary phase, and a mixture of 100 mmol·L-1 ammonium formate solution (containing 1% formic acid)and acetonitrile (15+85) was used as mobile phase. Diode array detector was used for determination. Linear relationships between values of peak area and mass concentration of D-iso-ascorbic acid, L-ascorbic acid and thiourea dioxide were kept in the same range of 1.0-100 mg·L-1, with the same detection limit (3S/N) of 1.0%. On the base of blank sample, test for recovery was made by standard addition method; values of recovery found were in the range of 94.7%-108%, with RSDs (n=6) of determined values less than 8.0%.
