    超声萃取-反相高效液相色谱-荧光法测定黑参中的苯并芘

    Determination of Benzopyrene in Black Ginseng by Ultrasonic Extraction-Reversed Phase High Performance Liquid Chromatography with Fluorescence Detector

    • 摘要: 0.500 0 g黑参样品用5 mL正己烷于40℃超声提取0.5 h,提取液用分子印迹固相萃取柱净化,净化液以Agilent TC-C18色谱柱为固定相,以88%(体积分数)乙腈溶液为流动相进行分离,采用反相高效液相色谱-荧光法测定。在优化的试验条件下,苯并芘的质量浓度在0.50~20.0 μg·L-1内与其峰面积呈线性关系,检出限(3S/N)为0.20 μg·kg-1。按标准加入法进行回收试验,回收率为92.8%~97.5%,相对标准偏差(n=6)均小于2.5%。

       

      Abstract: The sample of black ginseng (0.500 0 g) was extracted ultrasonically for 0.5 h at 40℃ with 5 mL of n-hexane. The extract was purified by molecularly imprinted solid phase extraction column, and then determined by reversed phase HPLC with fluorescence detector, using Agilent TC-C18 column as stationary phase and 80% (φ) acetonitrile solution as mobile phase for elution. Under the optimized test conditions, linearity relationship between values of peak area and mass concentration of benzopyrene was found in the range of 0.50-20.0 μg·L-1, with detection limit (3S/N) of 0.20 μg·kg-1. Test for recovery was made by standard addition method, giving values of recovery in the range of 92.8%-97.5% and RSDs (n=6) of the determined values all less than 2.5%.
