    柱前衍生化顶空-气相色谱-质谱法测定坚果和梅类食品中氰化物的含量

    Determination of Cyanide in Nuts and Plum Food by Headspace Gas Chromatography-Mass Spectrometry with Pre-Column Derivatization

    • 摘要: 提出了柱前衍生化顶空-气相色谱-质谱法测定坚果和梅类食品中氰化物(以CN-计,下同)含量的方法。取样品2.000 0 g,加1.0 g·L-1氢氧化钠溶液约60 mL,混匀,超声提取20 min,冷却至室温后用1.0 g·L-1氢氧化钠溶液定容至100 mL,过滤。取5 mL续滤液置于顶空瓶中,加入17%(体积分数)磷酸溶液200 μL,混合后加入10 g·L-1氯胺T溶液200 μL,于55 ℃衍生30 min。以DB-624UI毛细管色谱柱为固定相,采用气相色谱-质谱法测定所得溶液中氰化物的含量。结果显示:CN-的质量浓度在200 μg·L-1以内与对应的氰化物衍生物的峰面积呈线性关系,检出限(3S/N)为0.01 mg·kg-1。按照标准加入法进行回收试验,回收率为96.8%~103%,测定值的相对标准偏差(n=6)均小于4.0%。方法与国家标准方法GB 5009.36-2016中分光光度法进行对比,两种方法所得测定结果基本一致,无显著性差异。方法用于实际样品分析,结果显示坚果和梅类食品中都检出氰化物,建议将氰化物添加到GB 2762-2022的食品中污染物限量名录中,对其进行严格控制。

       

      Abstract: A method for determination of cyanide (calculated in CN-, the same below) in nuts and plum food by headspace gas chromatography-mass spectrometry with pre-column derivatization was proposed. The sample (2.000 0 g) was taken and mixed well with about 60 mL of 1.0 g·L-1 sodium hydroxide solution. The solution was extracted by ultrasonic for 20 min. After cooling to room temperature, the extract was made its volume up to 100 mL with 1.0 g·L-1 sodium hydroxide solution and filtered. 5 mL of the subsequent filtrate was put into the headspace bottle, and 200 μL of 17% (volume fraction) phosphoric acid solution was added. After mixing, 200 μL of 10 g·L-1 chloramine T solution was added and the solution was derivatized at 55 ℃ for 30 min. Cyanide in the obtained solution was determined by gas chromatography-mass spectrometry, with DB-624 UI capillary column as the stationary phase. As shown by the results, linear relationship between mass concentration of CN- and the corresponding peak area of cyanide derivative was found within 200 μg·L-1, with detection limit (3S/N) of 0.01 mg·kg-1. Test for recovery was made by the standard addition method, giving results in the range of 96.8%-103%, with RSDs (n=6) of the determined values less than 4.0%. This method was compared with the spectrophotometry according to the national standard method GB 5009.36-2016, and the determined results obtained by the two methods were basically consistent without significant difference. This method was used for analysis of the actual samples, and it was shown that cyanide was detected in all nuts and plum food, and it was recommended that cyanide should be added to the list of contaminant limits in the food of GB 2762-2022 for strict control.
