    4种谱图分析技术结合化学计量学在川麦冬产地区分中的应用比较

    Comparison of the Application of Four Spectra Analysis Techniques with Chemometrics in Production Area Differentiation of Sichuan Ophiopogon Japonicus

    • 摘要: 分别采用核磁共振氢谱(1H-NMR)、红外光谱(IR)、紫外光谱(UV)、高效液相色谱(HPLC)结合化学计量学对川麦冬产地进行区分,比较4种谱图分析技术的区分结果。获得11批不同产地川麦冬样品的1H-NMR、IR、UV图谱和HPLC指纹图谱,对其进行相似度评价,采用聚类分析、主成分分析和偏最小二乘-判别分析对样品进行分类。结果表明:来自四川省绵阳市三台县的8批样品的1H-NMR图谱和HPLC指纹图谱相似度均大于0.950,表现为极显著相关(P<0.01),可与其他产地样品较好地区分开来;IR图谱聚类分析将11批样品分为3类,而UV、1H-NMR图谱和HPLC指纹图谱聚类分析均无法细分产地;1H-NMR、IR图谱和HPLC指纹图谱偏最小二乘-判别分析结果一致,均将四川省绵阳市三台县的8批样品与其他产地的样品区分开。

       

      Abstract: Hydrogen magnetic resonance spectroscopy (1H-NMR), infrared spectroscopy (IR), ultraviolet spectroscopy (UV) and high performance liquid chromatography (HPLC) with chemometrics were used to distinguish the production area of Sichuan Ophiopogon japonicus, and the differentiation results of 4 spectral analysis techniques were compared. 1H-NMR, IR, UV spectra and HPLC fingerprints of 11 batch of Sichuan Ophiopogon japonicus samples from different production areas were obtained, and the similarity evaluation was carried out. Cluster analysis, principal component analysis and partial least square-discriminant analysis were used to classify the samples. As shown by the results, the similarities of 1H-NMR spectra and HPLC fingerprints of 8 batches of samples from Santai County, Mianyang City, Sichuan Province were greater than 0.950, showing a highly significant correlation (P<0.01), and that could be better distinguished from other production areas samples. The results of IR spectra with cluster analysis showed that 11 batches of samples could be divided into 3 categories, but those of UV, 1H-NMR spectra and HPLC fingerprint could not subdivide the production areas. The results of 1H-NMR, IR spectra and HPLC fingerprints with partial least square-discriminant analysis were consistent, that 8 batches of samples from Santai County, Mianyang City, Sichuan Province were distinguished from other production areas samples.
