    高效液相色谱指纹图谱结合化学计量学评价半夏泻心汤物质基准的质量

    Quality Evaluation of Banxia Xiexin Decoction Material Benchmark by High Performance Liqiud Chromatography Fingerprint with Chemometrics

    • 摘要: 为研究半夏泻心汤(BXD)物质基准的质量控制方法,进行了题示研究。以15批BXD物质基准为研究对象,建立高效液相色谱(HPLC)指纹图谱,与对照指纹图谱进行对比指认其中的共有峰,利用中药色谱指纹图谱相似度评价系统(2012.130723版本)进行相似度评价,采用薄层色谱法(TLC)定性鉴别15批BXD物质基准中人参,并结合聚类分析(CA)、主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA)等化学计量学方法寻找影响质量的主要差异成分。结果显示:建立了15批除人参以外的BXD物质基准HPLC指纹图谱,得到21个共有峰,指认出11个成分(腺苷、尿苷、鸟苷、黄芩苷、黄芩素、汉黄芩素、盐酸小檗碱、盐酸巴马汀、表小檗碱、6-姜辣素、甘草酸铵),相似度均大于0.900;对人参皂苷Rg1、Rb1、Re、Rf进行TLC定性鉴别,结果一致性良好。CA、PCA和OPLS-DA均将15批BXD物质基准分为4类,PCA得到3个主成分,累计方差贡献率为85.598%,OPLS-DA得到12个对BXD物质基准整体质量贡献率大的差异成分,包含汉黄芩素、盐酸巴马汀、甘草酸铵和9种未知物,分别归属于黄芩、黄连和甘草等,进而说明上述3种饮片对BXD物质基准的质量影响较大。

       

      Abstract: To investigate the quality control method for banxia xiexin decoction (BXD) material benchmark, the title study was conducted. With 15 batches of BXD material benchmarks as research object, high performance liquid chromatography (HPLC) fingerprints were established, and common peaks were identified by comparing with reference fingerprint. Similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine (2012.130723 edition) was used for similarity evaluation. Ginsengs in 15 batches of BXD material benchmarks were qualitatively identified by thin layer chromatography (TLC). Chemometrics methods including cluster analysis (CA), principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) were used to find the main difference components affecting the quality. It was shown that HPLC fingerprints of 15 batches of BXD material benchmarks except ginsengs were established, and 21 common peaks were obtained. And 11 components (adenosine, uridine, guanosine, baicalin, baicalein, wogonin, berberine hydrochloride, palmatine hydrochloride, epiberberine, 6-gingerol and ammonium glycyrrhizinate) were identified, and the similarity was greater than 0. 900. Qualitative identification by TLC of ginsenosides Rg1, Rb1, Re and Rf were shown good consistency. 15 batches of BXD material benchmarks were divided into 4 categories by CA, PCA and OPLS-DA. 3 principal components were obtained by PCA, with cumulative variance contribution rate of 85. 598%. 12 difference components with large contribution rate to overall quality of BXD material benchmarks were obtained by OPLS-DA, including wogonin, palmatine hydrochloride, ammonium glycyrrhizinate and 9 unknown substances, respectively belonging to scutellaria baicalensis, rhizoma coptis and liquorice, which were indicated that the above 3 kinds of decoction pieces had a great effect on quality of BXD material benchmarks.
