    单糖乙酰化条件的优化

    Optimization of Conditions for Acetylation of Monosaccharides

    • 摘要: 利用气相色谱-质谱联用技术(GC-MS)检测5种常见单糖(木糖、阿拉伯糖、甘露糖、葡萄糖、半乳糖)的乙酰化产物含量.为在预衍生化中取得较高的效率,采用正交试验综合考察了还原时间、还原温度、乙酰化温度、乙酰化时间等因素对单糖回收率的影响及规律.结果表明,影响最为显著的因子为还原温度和乙酰化时间.总结出了单糖衍生化最佳的反应条件为还原温度50 ℃、还原时间2.5 h;乙酰化温度90 ℃、乙酰化时间2.5 h.

       

      Abstract: Hyphenated GC and MS was used in the determination of acetylation products of 5 common monosaccharides,i.e.,xylose,arabinose,mannose,glucose and galactose.For obtaining high efficiency of pre-derivatization,conditions for acetylation,including the time and temperature of reduction,time and temperature of acetylation and etc.,were studied and optimized by using the orthogonal experimental designing method.It was found that the temperature of reduction and time of acetylation were found to be the significant factors influential to the recovery of monosaccharides.The optimized acetylation conditions were found as follows:① temperature of reduction:50 ℃;② time of reduction:2.5 h;③ temperature of acetylation:90 ℃ and ④ time of acetylation 2.5 h.
