    免疫共振散射光谱法测定痕量人血清前白蛋白

    Immuno Resonance Scattering Spectrometric Determination of Traces of Human Serum Pre-albumin

    • 摘要: 在pH 7.6的磷酸氢二钠-磷酸二氢钠缓冲溶液中,羊抗人前白蛋白与前白蛋白发生特异性结合生成免疫复合物,导致体系的共振散射增强。体系在最大散射波长470 nm处,前白蛋白的质量浓度在0.067~2.82 mg·L-1范围内与共振散射增强强度(ΔI)呈线性关系,检出限(3S/N)为0.028 mg·L-1。应用于测定人血清前白蛋白,测定值相对标准偏差(n=6)在1.15%~3.00%之间。

       

      Abstract: In a PBS of pH 7.6, enhancement of intensity of resonance scattering due to specific binding to form immune complex between goat anti-human pre-albumin and pre-albumin was observed. The magnitude of increase in resonance scattering intensity (ΔI) was found to keep linear relationship with the mass concentration of pre-albumin in the range of 0.067-2.82 mg·L-1 when measured at 470 nm (the maximum scattering wavelength of the reaction system). Detection limit (3S/N) of the method was found to be 0.028 mg·L-1. In its application to determination of human serum pre-albumin, values of RSD′s (n=6) found were in the range of 1.15%-3.00%.
