    荧光分光光度法研究对乙酰氨基酚与牛血清白蛋白之间的相互作用

    Fluorospectrophotometric Study on the Interaction Between Paracetanol and Bovine Serum Albumin

    • 摘要: 在模拟人体生理环境下,用荧光分光光度法及紫外-可见分光光度法研究了解热镇痛药对乙酰氨基酚(简写作PTL)与牛血清蛋白(BSA)之间的相互反应.试验结果发现:由于非辐射能量转移引起的荧光猝灭,BSA的荧光强度因与PTL相互之间的结合反应而明显降低.按Stern-Volmer方程和Lineweaver-Burk方程对所得实际数据进行了处理,求得此结合反应的静态平衡常数(KLB)为2.592×103mol·L-1(297 K),其结合位置距212位色氨酸1.94 nm.根据结合反应的热力学参数,推断PTL与BSA之间的结合力为疏水作用力.

       

      Abstract: The binding reaction of the drug,paracetanol (PTL),with bovine serum albumin (BSA) was studied by fluorescence spectrophotometry and UV-VIS spectrophotometry,in simulating physiological environment of human body.It was found that the fluorescence intensity of BSA was remarkably reduced due to fluorescence quenching through non-radiative energy transfer.Experimental data were treated by the stern-Volmer equation and the Lineweaver-Burk equation,and the static equilibrium constant (KLB) of the binding reaction was found to be 2.592×103mol·L-1 (297 K),and the binding locality was 1.94 nm apart from the 212 sited tryptophan.According to the thermodynamic parameters of the reaction,the binding power was primarily the force due to hydrophobic action.
