    高效液相色谱法测定四环素类抗生药物中土霉素、强力霉素、四环素和金霉素

    Determination of Oxytetracycline,Doxycycline,Tetracycline and Chlortetracycline in Tetracycline Antibiotics by HPLC

    • 摘要: 利用高效液相色谱,发展了一种快速、灵敏、同时测定土霉素、强力霉素、四环素和金霉素的方法.在反相C18柱上进行梯度洗脱分离,流动相由甲醇和乙酸钠缓冲溶液组成(内含EDTA和氯化钙,pH 8.10),紫外检测波长为386 nm.四环素类药物的质量浓度在8~4 000 μg·L-1范围内呈线性关系,回收率为95%~102%,相对标准偏差为1.2%~3.6%,检出限分别为6,13,6和7 μg·L-1,方法应用于四环素类抗生药物的分析.

       

      Abstract: A rapid and sensitive high-performance liquid chromatographic method for the simultaneous determination of oxytetracycline,doxycycline,tetracycline and chlortetracycline was developed. The separation was performed on a reverse-phase C18 column with gradient elution using a mixed solution composed of methanol and sodium acetate buffer (containing disodium salt of EDTA and calcium chloride,pH 8.10). UV detection at 386 nm was used,linearity of the calibration curve was obtained in the range of 8-4 000 μg·L-1. The detection limits for oxytetracycline,doxycycline,tetracycline and chlortetracycline were 6,13,6 and 7 μg·L-1 respectively. The method has been applied to the analysis of commercial tetracycline antibiotics,giving RSD′s in the range from 1.2% to 3.6%,and recoveries from 95% to 102%.
