    流动注射化学发光法测定葛根黄酮

    Determination of Pueraria Flavonoid by Flow Injection Chemiluminescence Method

    • 摘要: 基于碱性条件下,葛根黄酮对鲁米诺-过氧化氢化学发光体系有显著的抑制作用,建立了测定葛根黄酮的流动注射化学发光法。在已优化的条件下,葛根黄酮的质量浓度在0.50~30.0 mg·L-1范围内与其化学发光强度的改变值ΔI呈线性关系,方法的检出限(3S/N)为0.08 mg·L-1。该方法用于葛根中葛根黄酮的测定,结果与紫外分光光度法的测定结果一致。加标回收率在96.4%~102%之间,测定值的相对标准偏差(n=5)在2.9%~3.5%之间。

       

      Abstract: A method of flow injection chemiluminescence was proposed for the determination of pueraria flavonoid, based on the decreasing effect of pueraria flavonoid on Luminol-H2O2 chemiluminescence system. Under the optimum conditions, linear relationship was found between the decrease of chemiluminescence intensity and the mass concentration of pueraria flavonoid in the range of 0.50-30.0 mg·L-1, with detection limit of 0.08 mg·L-1. The method was applied for the determination of pueraria flavonoid in pueraria lobata, giving results in agreement with that obtained by UV spectrophotometry, with RSD (n=5) in the range of 2.9%-3.5%. The recovery rates measured by standard addition method were in the range of 96.4%-102%.
