柱后衍生高效液相色谱法测定罗汉果甜甙中黄曲霉毒素B1
HPLC Determination of Aflatoxin B1 in Fructoside of Fructus Momordicae with Post-Column Derivation

摘要: 用柱后衍生高效液相色谱法测定了罗汉果甜甙中黄曲霉毒素B1(AFX B1).用MYCOTOXTM C18作为反相色谱柱,流动相为由甲醇、乙腈及水以22+22+56之比例混合的混合液.柱后衍生试剂为100 mg·L-1碘的水溶液,以荧光检测器在发射波长为365 nm及激发波长为430 nm条件下进行测定.在AFX B1的峰面积与质量浓度(mg·L-1)之间呈线性关系,其回归方程为y=14.222 x+0.625 3(r=0.998),在三个浓度水平(5.1,40.8及102 μg·kg-1)上进行回收率及精密度试验,所得回收率在81.4%~94.2%之间,相对标准偏差(n=10)在2.1%~4.5%之间.Abstract: Aflatoxin B1 (AFX B1) in fructoside of fructus momordicae was determined by HPLC with pos-column derivatization.MYCOTOXTM C18 was used as reversed-phase chromatographic column (4.6 mm×250 mm),and a mixture containing methanol,acetonitrile and water in the ratio of 22+22+56 was used as mobile phase.A water solution of iodine (100 mg·L-1) was used the derivatizing reagent.Measurement was made by fluorescence detector with λex 365 nm and λem 430 nm.Linear relationship between peak area and mass concentration (mg·L-1) was shown by the linear regression equation,y=14.222 x+0.625 3 (r=0.998).Tests for recovery and precision were performed at 3 different concentration levels (5.1,40.8 and 102 μg·kg-1 of AFX B1),and values of recovery and RSD′s (n=10) obtained were in the ranges of 81.4%-94.2% and 2.1%-4.5% respectively.
