    反相高效液相色谱法测定花椒的3种黄酮苷元的含量

    RP-HPLC Determination of 3 Flavonoid Aglycones of Zanthoxylum Bungeanum

    • 摘要: 提出了反相高效液相色谱法测定花椒的3种水解黄酮苷元槲皮素、山柰酚、异鼠李素的含量.采用Zorbax Eclipse C18色谱柱(4.6 mm×150 mm,5 μm),以甲醇-0.4%(体积分数)磷酸溶液(1+1)混合溶液为流动相,在360 nm波长处进行测定.槲皮素的质量浓度在2.19~109.6 mg·L-1,山柰酚的质量浓度在2.28~114.0 mg·L-1,异鼠李素的质量浓度在2.80~140.0 mg·L-1时分别与其峰面积呈线性关系.槲皮素、山柰酚和异鼠李素的加标回收率分别为95.6%,104.4%,103.8%,相对标准偏差(n=5)分别为2.98%,3.97%,4.30%.

       

      Abstract: RP-HPLC was applied to the determination of 3 flavonoid aglycones,including quercetin,kaempferol and isorhamnetin in zanthoxylum bungeanum.The sample solution was separated by using Zorbax Eclipse C18 column (4.6 mm×150 mm,5 μm) as stationary phase and a mixture of methanol and 0.4% (φ) phosphoric acid solution (mixed 1 to 1 by vol.) as mobile phase.The contents of the 3 flavonoid aglycones were determined at the wavelength 360 nm.Linear relationships between values of peak area and mass concentration of quercetin,kaempferol and isorhamnetin were kept in ranges of 2.19-109.6 mg·L-1,2.28-114.0 mg·L-1 and 2.80-140.0 mg·L-1,respectively.Tests for recovery and precision were made by standard addition method.Values of recovery and RSD′s (n=5) found were 95.6% and 2.98% for quercetin,104.4% and 3.97% for kaempferol,103.8% and 4.30% for isorhamnetin,respectively.
