    毛细管区带电泳法同时测定茶叶中5种活性成分的含量

    Simultaneous Determination of 5 Active Components in Tea by Capillary Zone Electrophoresis

    • 摘要: 提出了毛细管区带电泳法同时测定茶叶中儿茶素、表儿茶素、原儿茶酸、原儿茶醛和槲皮素等5种化合物含量的方法。采用无涂层适应毛细管柱(57 cm×75 μm,有效长度50 cm)分离,以含10 mmol·L-1四硼酸钠、5 mmol·L-1 β-环糊精、15%(体积分数,下同)乙腈、2%乙二醇pH 9.11的缓冲溶液为电泳介质,分离电压为20 kV。儿茶素、表儿茶素、原儿茶酸、原儿茶醛和槲皮素线性范围分别为8.152 5~217.4,8.152 5~217.4,2.174~217.4,1.739~150.0,1.739~150.0 mg·L-1。方法用于不同茶叶提取物样品中5种化合物含量的测定,测得平均回收率在94.6%~107%之间,测定值的相对标准偏差(n=5)在0.17%~4.4%之间。

       

      Abstract: Capillary zone electrophoresis (CZE) was applied to the determination of 5 active components (i.e., protocatechuic aldehyde, protocatechic acid, epicatechin, DL-catechin and quercitin) in tea. Efficient separation was attained by using the un-coated adaptive quartz capillary column (57 cm×75 μm, effective length 50 cm) and the electrophoretic medium of a buffer mixture, containing 10 mmol·L-1 Na2B4O7, 5 mmol·L-1 β-CD, 15% (φ) acetonitrile and 2% (φ) ethylene glycol (acidity adjusted to pH 9.11) with separation voltage of 20 kV. Linearity ranges were found between 8.152 5-217.4 mg·L-1 (for protocatechuic aldehyde), 8.152 5-217.4 mg·L-1 (for protocatechic acid), 2.174-217.4 mg·L-1 (for epicatechin), 1.739-150.0 mg·L-1 (for DL-catechin) and 1.739-150.0 mg·L-1 (for quercitin). The proposed method was used for determination of the 5 components in tea samples, giving values of recovery and RSD′s (n=5) in the ranges of 94.6%-107% and 0.17%-4.4% respectively.
