    高效液相色谱法测定对虾中3种磺胺类药物残留量

    HPLC Determination of Residual Amounts of 3 Sulfonamides in Prawn

    • 摘要: 提出了高效液相色谱法测定对虾中3种磺胺类药物磺胺嘧啶(SD)、磺胺吡啶(SPD)和磺胺噻唑(ST)的残留量。样品经乙腈提取,正己烷脱脂净化,高效液相色谱分离,紫外检测器检测,外标法定量。以Hypersil ODS色谱柱(250 mm×4.0 mm,5 μm)为分离柱,以甲醇和水以体积比25比75的混合液为流动相。3种磺胺类药物的质量浓度与其峰面积呈线性关系。磺胺嘧啶、磺胺吡啶和磺胺噻唑的回收率分别为93.2%,89.9%,88.7%;相对标准偏差(n=6)依次为3.1%,0.1%,1.1%。

       

      Abstract: HPLC was applied to the determinations of 3 sulfonamides in prawn, including sulfadiazine (SD), sulfapyridine (SPD) and sulfathiazole (ST). The sample was extracted by acetonitrile, and the extract obtained was purified by defatting with n-hexane. The Hypersil ODS column (250 mm×4.0 mm, 5 μm) was used as stationary phase, and a mixed solution of methanol and water (mixed in the a ratio of 25 to 75) was used as mobile phase. The eluate from column was used for HPLC analysis with UV detector. External standard calibration was adopted in the determination. Linear relationships in definite ranges between the values of peak area and mass concentration of the 3 sulfonamides were obtained. The values of RSD′s (n=6) and recovery found by standard addition method were 93.0% and 3.1% for sulfadiazine , 89.9% and 0.1% for sulfapyridine, 88.7% and 1.1% for sulfathiazole respectively.
