高效液相色谱法测定植物果实中儿茶素、表儿茶素和原花青素B2
HPLC Determination of Catechin, Epicatechin and Procyanidin B2 in Plant Fruits

摘要: 提出了高效液相色谱法测定植物果实中儿茶素、表儿茶素和原花青素B2。样品经乙醇(6+4)溶液索氏提取后,用固相萃取技术对提取液进行净化。以SinoChrom ODS C18色谱柱为分离柱,以乙腈-0.1%(φ)磷酸溶液(15+85)为流动相进行洗脱,检测波长为280 nm。3种目标分析物质量浓度的线性范围为5.0~100.0 mg·L-1,方法的检出限(3S/N)在1.03~2.38 mg·L-1之间。对样品进行加标回收试验,回收率在83.6%~98.5%之间,测定值的相对标准偏差(n=3)在0.4%~2.1%之间。Abstract: HPLC was applied for the determination of catechin, epicatechin and procyanidin B2 in plant fruits. The sample was extracted with ethanol solution (6+4) by soxhlet extraction. The extract was purified by solid phase extraction and then was separated on a SinoChrom ODS C18 column with a mixture of acetonitrile and 0.1% (φ) phosphoric acid solution as mobile phase. UV absorbance was measured at 280 nm. The linearity range of the mass concentration of the 3 target analytes was 5.0-100.0 mg·L-1, with detection limits (3S/N) in the range of 1.03-2.38 mg·L-1. The recovery rates measured by standard addition method were in the range of 83.6%-98.5% with RSD (n=3) in the range of 0.4%-2.1%.
