    高效液相色谱法同时测定黄花鱼中4种禁用黄色工业染料

    Simultaneous HPLC Determination of 4 Contraband Yellow Industrial Dyes in Yellow Croaker

    • 摘要: 采用高效液相色谱法同时测定黄花鱼中4种工业染料(碱性嫩黄O、橙黄G、碱性橙Ⅱ和皂黄)。以Kromasil C18柱为分离柱,以不同体积比的甲醇、水和20 mmol·L-1乙酸铵溶液的混合液为流动相进行梯度洗脱,在波长430 nm处进行测定。上述4种染料的线性范围依次为0.05~2.0 mg·L-1,1.0~15.0 mg·L-1,0.1~5.0 mg·L-1及0.4~15.0 mg·L-1。方法的检出限(3S/N)在0.002 5~0.05 μg·kg-1之间。以黄花鱼样品为基体进行回收试验,测得回收率在96.4%~98.1%之间,测定值的相对标准偏差(n=6)在1.3%~2.5%之间。

       

      Abstract: HPLC was applied to the determination of 4 contraband yellow industrial dyes, i.e. auramine O, orange G, basic orange Ⅱ and metanil yellow, in yellow croaker. Kromasil C18 column was used as stationary phase for separation, and mixtures of methanol, water and 20 mmol·L-1 ammonium acetate solution in different ratios were used as mobile phase in gradient elution. UV-detection at 430 nm was adopted in the determination. Linearity ranges for the 4 above mentioned dyes were found between 0.05-2.0 mg·L-1, 1.0-15.0 mg·L-1, 0.1-5.0 mg·L-1 and 0.4-15.0 mg·L-1 respectively. Values of detection limit (3S/N) of the dyes were found in the range of 0.002 5-0.05 μg·kg-1. On the base of a yellow croaker sample, test for recovery was made by standard addition method, values of recovery found were in the range of 96.4%-98.1%, with RSD′s (n=6) in the range of 1.3%-2.5%.
