    氢化物发生-原子吸收光谱法测定黄芪中硒

    HG-AAS Deterimination of Selenium in Astragalus

    • 摘要: 黄芪试样经硝酸-过氧化氢消解,用氢化物发生-原子吸收光谱法测定黄芪中总硒、无机硒和有机硒的含量。使用15 g·L-1硼氢化钾-3 g·L-1氢氧化钠溶液与溶液中硒离子反应生成氢化物。分析中采用载气流量240 mL·min-1。硒的质量浓度在60 μg·L-1以内与吸光度呈线性关系,方法的检出限(3σ)为0.21 μg·L-1。应用此法测定黄芪中硒的含量,总硒测定值为105 μg·g-1、硒(Ⅳ)测定值为15 μg·g-1、硒(Ⅵ)测定值为9 μg·g-1、有机硒测定值为81 μg·g-1。黄芪中的硒主要存在形式为有机硒,占总硒含量的77%。

       

      Abstract: The sample of astragalus was digested with HNO3 and H2O2, and the contents of total selenium, inorganic selenium and organic selenium were determined by HG-AAS. Hydride of Se ion was generated by reacting with 15 g·L-1 KBH4-3 g·L-1 NaOH solution. Flow-rate of 240 mL·min-1 for carrier gas was taken in the analysis. Linear relationship between values of absorbance and mass concentration of Se found was in the range within 60 μg·L-1, with detection limit (3σ) of 0.21 μg·L-1. The proposed method was applied to the determination of Se in astragalus, giving values of 105 μg·g-1 for total Se, 15 μg·g-1 for Se(Ⅳ), 9 μg·g-1 for Se(Ⅵ) and 81 μg·g-1 for organic Se. It was found that the main species of Se in astragalus was organic Se, amounting to 77% of the total Se.
