    5-Br-PADAP双波长分光光度法测定酒样中甲醇

    DW-Spectrophotometric Determination of Methanol in Wine with Reagent 5-Br-PADAP

    • 摘要: 用2-(5-溴-2-吡啶偶氮)-5-二乙氨基酚(简写作5-Br-PADAP)作为在乙醇溶液(如酒)中识别甲醇的试剂,在pH 9.0的乙醇溶液中,5-Br-PADAP的吸收峰位于444 nm处,而有甲醇存在时,在552 nm处出现新的吸收峰,即吸收峰发生红移,两吸收峰之间相差108 nm,从有甲醇存在的5-Br-PADAP乙醇溶液的吸收光谱上可见到在444 nm及552 nm两波长处分别有负吸收峰和正吸收峰,在两处的吸光度与甲醇浓度之间呈线性关系,基于以上事实,提出了测定酒中甲醇量的双波长分光光度法,以552 nm为测定波长,444 nm为参比波长,所提出的方法用于测定假冒酒及工业酒精试样中甲醇的含量,测定结果的相对标准偏差(n=8)均小于1%.用标准加入法测得的回收率在98.7%~100.1%之间,对方法所涉及的反应机理也作了简要的讨论.

       

      Abstract: 2-(5-Bromo-2-pyridylazo)-5-diethylaminophenol (5-Br-PADAP) was used as a reagent to recognize the presence of methanol in an ethanolic solution,such as wine.In an ethanolic solution of pH 9.0,absorption maximum (λmax) of 5-Br-PADAP was found at 444 nm and when methanol was present in the solution,the absorption maximum was bathochromically shifted to 552 nm,making a difference of 108 nm in wavelength between the 2 absorption maxima.It was observed in the absorption spectra of ethanolic solution 5-Br-PADAP in the presence of methanol,positive and negative absorption maxima were found at 552 nm and 444 nm respectively,and the values of absorbance,measured at either of the 2 wavelength,were correlated linearly with the concentration of methanol present in the solution.It was proposed that based on facts mentioned above DW-measurement was applied to the determination of methanol in wine,selecting the wavelength 552 nm as the wavelength of determination and 444 nm as the reference wavelength.The proposed method was applied to the determination of methanol in samples of imitated wine and industrial alcohol,giving values of RSD′s (n=8) less than 1%,and values of recovery in the range of 98.7%-100.1%.A discussion on the raction mechanism was also given.
