高效液相色谱流动相手性添加剂法拆分吲达帕胺对映体
Resolution of Enantiomers of Indapamide by HPLC with Mobile Phase Added with Chiral Agent

摘要: 以β-环糊精(β-CD)作为手性流动相添加剂,应用反相高效液相色谱研究吲达帕胺对映体的拆分.在Kromasil C18色谱柱上,采用含12.0 mmol·L-1 β-CD,0.05 mol·L-1磷酸二氢钠、2%(体积分数)的三乙胺、pH 2.5的混合溶液-甲醇(75+25)为流动相,流量为0.8 mL·min-1,室温下拆分,吲达帕胺对映体可得到良好分离.Abstract: β-Cyclodextrin (12.0 mmol·L-1) was used as an additive added to a solution containing 0.05 mol·L-1 NaH2PO4 and 2% (by vol.) of triethylamine (pH 2.5),which is mixed with methanol in the ratio of 75∶25 and used as the mobile phase in the resolution of enantiomers of indapamide by RP-HPLC. The Kromasil C18 column was used as chromatographic column. Flow-rate of 0.8 mL·min-1 was taken for the mobile phase. Under the optimun conditions,the enantiomers of iodapamide were satisfactorily under room temperature.
