分光光度法测定葛根中总黄酮含量
Spectrophotometric Determination of Total Flavones in Root of Kudzu Vine

摘要: 采用分光光度法测定葛根中总黄酮的含量。样品经乙醚浸泡去除杂质,用甲醇超声波提取。以芦丁为标准品制作标准曲线,于最大吸收波长510 nm处测定。芦丁的质量浓度在8.48~28.92 mg·L-1范围内与其吸光度呈线性关系,方法的检出限(3S/N)为0.75 mg·L-1。方法用于葛根样品分析,测得总黄酮平均含量为1.64 mg·g-1,测定值的相对标准偏差(n=6)为1.2%。用标准加入法做回收试验,测得回收率在97.2%~101%之间。Abstract: A spectrophotometric method for determination of total flavones in root of kudzu vine was proposed. The sample was soaked with ether to remove impurities, and extracted ultrasonically with methanol. Standard curve was prepared with rutin as reference standard, linear relationship between values of absorbance and mass concentration was found in the range of 8.48-28.92 mg·L-1 as measured at the absorption maximum of 510 nm. Detection limit (3S/N) of 0.75 mg ·L-1 was obtained. The proposed method was applied to the determination of total flavones in a sample of root of kudzu vine, giving average content 1.64 mg·g-1 and RSD (n=6) of 1.2%. Recovery was tested by standard addition method, values of recovery were found in the range of 97.2%-101%.
