    高效液相色谱法测定植物油中麦固醇的含量

    HPLC Determination of Sitosterol in Vegetable Oil

    • 摘要: 采用高效液相色谱法测定植物油中麦固醇的含量。试验优化了提取油脂混合物添加的麦固醇的工艺因素:最佳提取次数为4次;离心转速为2 000 r·min-1;皂化温度为80 ℃;皂化时间为3.5 h。用C18反相色谱柱为分离柱,以甲醇-水(98.5+1.5)溶液为流动相,检测波长为210 nm,麦固醇的质量浓度在12.5~1 000 mg·L-1范围内与峰面积呈线性关系,方法的检出限(3S/N)为5 mg·L-1。加标回收率在86.2%~98.7%之间,测定值的相对标准偏差(n=6)在2.4%~3.9%之间。

       

      Abstract: HPLC was applied to the determination of sitosterol in vegetable oil. The extraction conditions of additional sitosterol in the mixed oil were optimized as follows: the extraction times of 4 times; the centrifugal speed of 2 000 r·min-1; the saponification temperature at 80 ℃ for 3.5 h. C18 reverse-phase chromatographic column was used as stationary phase and a mixture of methanol and water (98.5+1.5) was used as mobile phase. UV-detection was made at the wavelength of 210 nm. Linear relationship between values of peak area and mass concentration of sitosterol was kept in the range of 12.5-1 000 mg·L-1 with detection limit (3S/N) of 5 mg·L-1. Values of recovery found by standard addition method were in the range of 86.2%-98.7%, and values of RSD′s (n=6) in the range of 2.4%-3.9%.
