    固相萃取-高效液相色谱法测定栀子中芦丁含量

    HPLC Determination of Rutin in Gardenia with Solid Phase Extraction

    • 摘要: 提出了固相萃取-高效液相色谱法测定栀子中芦丁含量的方法。样品以甲醇-水(80+20)混合液提取,C18固相萃取小柱净化,所得净化液采用C18色谱柱为分离柱,以乙腈、0.2 mmol·L-1硫酸铜溶液、三乙胺、乙酸以体积比30比70比0.5比0.7组成的混合液为流动相,在检测波长360 nm处进行测定。芦丁的质量浓度在4.65~46.5 mg·L-1范围内与峰面积呈线性关系,方法检出限(3S/N)为0.86 mg·L-1。加标回收率在95.6%~97.5%之间,测定值的相对标准偏差(n=6)均小于1.5%。

       

      Abstract: A method of HPLC for the determination of rutin in gardenia with solid phase extraction was proposed. The sample was extracted with a mixture of methanol and water (80+20) and purified by passing through C18-solid phase extraction column. C18 chromatographic column was used as stationary phase, and a mixture of acetonitile, 0.2 mmol·L-1 CuSO4 solution , triethylamine and acetic acid (30+70+0.5+0.7) was used as mobile phase. UV-detection at 360 nm was adopted in the determination. Linear relationship between values of peak area and mass concentration of rutin was kept in the range of 4.65-46.5 mg·L-1, with detection limit (3S/N) of 0.86 mg·L-1. Values of recovery found by standard addition method were in the range of 95.6%-97.5%, and values of RSD′s (n=6) less than 1.5%.
