基于血红蛋白/金纳米粒子/聚二烯丙基二甲基氯化铵-多壁碳纳米管生物传感器测定过氧化氢
Application of the Biosensor Hb/AuNP′s/PDDA-MWCNT′s/GCE to Determination of Hydrogen Peroxide

摘要: 将分散于聚二烯丙基二甲基氯化铵(PDDA)中的多壁碳纳米管(MWCNT′s)滴涂在玻碳电极(GCE)表面制成PDDA-MWCNT′s/GCE修饰电极(简称电极Ⅰ);在氯金酸溶液中用恒电位沉积法使金纳米颗粒(AuNP′s)积镀于电极Ⅰ表面,制得AuNP′s/PDDA-MWNCT′s/GCE修饰电极(简称电极Ⅱ);将血红蛋白(Hb)滴于电极Ⅱ表面,用磷酸盐缓冲溶液冲洗后制成可供测定过氧化氢用的生物传感器Hb/AuNP′s/PDDA-MWCNT′s/GCE(简称电极Ⅲ)。与裸GCE、电极Ⅰ和Ⅱ相比较,在pH 6.8的磷酸盐缓冲溶液中,过氧化氢在电极Ⅲ上的还原峰电流明显提高,其值与过氧化氢浓度在1.0~1 800 μmol·L-1范围内呈线性关系,其检出限(3S/N)为0.8 μmol·L-1。应用此生物传感器测定消毒液中过氧化氢的含量,测定值与高锰酸钾滴定法的测定值相符。Abstract: A dispersion of multi-wall carbon nanotubes (MWCNT′s) in polydipropylenedimethylammonium chloride (PDDA) was added dropwise onto the surface of glassy carbon electrode (GCE) to prepare modified electrode of PDDA-MWCNT′s/GCE (abbr. as electrode Ⅰ); gold nanoparticles (AuNP′s) were plated on the surface of electrode Ⅰ by constant potential electro-deposition, the modified electrode of AuNP′s/PDDA-MWCNT′s/GCE (abbr. as electrode Ⅱ) was obtained. Haemoglobin (Hb) was dropped and immobilized on the surface of electrode Ⅱ to obtain the biosensor Hb/AuNP′s/PDDA-MWCNT′s/GCE (abbr. as electrode Ⅲ) which after rinsing with PBS of pH 6.8, was used for determination of H2O2. As compared with bare GCE, electrode Ⅰ and electrode Ⅱ, in a PBS of pH 6.8, the reduction peak current of H2O2 at electrode Ⅲ was significantly increased. Linear relationship was kept between values of reduction peak current and concentration of H2O2 in the range of 1.0 to 1 800 μmol·L-1, with detection limit (3S/N) of 0.8 μmol·L-1. In the application of this biosensor to the determination of H2O2 in a disinfectant, results obtained by this method were in consistency with those found by the KMnO4 titrimetric method.
