索氏提取-反相高效液相色谱法测定人工晶状体中甲基丙烯酸甲酯单体残留量
RP-HPLC Determination of Residual Amount of Methyl Methacrylate Monomers in Intraocular lens by Soxhlet′s Extraction

摘要: 应用反相高效液相色谱法测定了由聚甲基丙烯酸甲酯(PMMA)或由亲水性丙烯酸酯(HMA)作原材料制成的人工晶状体(IOL)中甲基丙烯酸甲酯单体(MMA)的残留量。采用索氏提取法分离MMA,选用异丙醇作为PMMA制成的IOL样品的提取溶剂,而选用乙腈作为HMA制成的IOL样品的提取溶剂。萃取液采用旋转蒸发仪浓缩,浓缩液经Waters C18色谱柱分离,用乙腈-水(30+70)溶液洗脱,在波长220 nm处进行紫外检测。MMA的质量浓度在0.33~42.08 mg·L-1范围内与峰面积呈线性关系,方法的检出限(3S/N)为0.02 mg·L-1。日内和日间相对标准偏差(n=6)分别为0.72%和0.92%,回收率在98.5%~106.2%之间。Abstract: RP-HPLC was applied to the determination of residual amount of methyl methacrylate (MMA) monomer in intraocular lens (IOL) made of polymethyl methacrylate (PMMA) or made of hydrophilic methacrylate (HMA). Soxhlet′s extraction was used for the separation of MMA monomer; isopropanol was selected as extraction solvent for IOL sample of PMMA and acetonitrile was selected as extraction solvent for IOL sample of HMA. The extracts were concentrated by rotary evaporator, and the concentrate was used for RP-HPLC analysis. Waters C18 column was used as stationary phase and acetonitrile-H2O mixture (30+70) was used as mobile phase in the separation. Ultraviolet detector at 220 nm was adopted in the determination. Linear relationship between values of peak area and mass concentration of MMA was obtained in the range of 0.33-42.08 mg·L-1 with detection limit (3S/N) 0.02 mg·L-1. Values of recovery were ranged from 98.5% to 106.2%, and RSD′s (n=6) of intra-day and inter-day found were 0.72% and 0.92% respectively.
