    响应面分析法和单因素分析法优化艾草中黄酮类物质的提取条件

    Optimization of Conditions of Extraction of Flavonoids from Wormwood with the Algorithms of Response Surface Analysis and Single Factor Analysis

    • 摘要: 选择分子荧光光度法对艾草中黄酮类物质的提取条件的优化试验试液中的黄酮类物质进行测定,从而算得不同条件下所达到的提取率以确定最佳的提取条件。研究了提取过程中的三因素:乙醇溶液体积分数、超声时间和艾草质量与乙醇溶液体积的比值。并按单因素分析法测得提取过程中3项因素最佳参数分别为:乙醇溶液的体积分数为50%,超声提取的时间为40 min,艾草样品和50%乙醇溶液的比值为1.0 g比15 mL。在此条件下,艾草中黄酮类物质的提取率为1.07%。以上述3项因素作为响应面分析法的优化因素,利用Design Expert 8.0软件进行三因素三水平的析因试验,对此提取条件作进一步的优化,得到了最佳的提取条件为:①乙醇溶液的体积分数为49.96%;②超声提取时间为30 min;③艾草样品与50%乙醇溶液的比值为0.79 g比15 mL。在此条件下提取率可达1.09%。按此条件称取艾草粉末样品0.790 0 g (3份),分别按上述优化的条件进行超声提取后,经过滤并离心后取其上清液(分取量应按样品中黄酮类物质的估计含量决定,使其量落在标准曲线的线性范围0.006~0.018 g·L-1之间)1 mL,依次加入5.3 g·L-1硝酸铝溶液2.38 mL和乙酸盐缓冲溶液(pH 5.6)0.9 mL后,加入50%乙醇溶液将其体积定容至10.0 mL。待黄酮类物质与铝(Ⅲ)离子的荧光配位反应完全后,测定其荧光发射强度。此3份样品的黄酮类物质平均提取率为1.08%,与理论预测值的相对误差为-0.92%。

       

      Abstract: Molecular fluorescence photometry was applied to the determination of flavonoids in test solutions obtained from the study on optimization of conditions of extraction of flavonoids from wormwood, and the optimum conditions of the extraction were determined on the base of the data of rate of extraction found. 3 factors, i.e., volume fraction of the ethanol solution, time for the ultrasonic extraction and ratio of mass of wormwood sample to volume of the ethanol solution used for extraction, were studied. As found by single factor analysis, values of the 3 factors were obtained as follows:① volume fraction of ethanol solution:50%; ② time for ultrosonic extraction:40 min and ③ ratio of mass of wormwood sample to volume of 50% C2H5OH solution:1.0 g to 15 mL. Rate of extraction attaining to a value of 1.07% was obtained under this condition. The 3 factors were taken as the factors to be optimized furthermore by the response surface analysis using the Design Expert 8.0 software by testing at factorial design of 3 factors and 3 levels. The optimum values of the 3 factors found by this method were as follows:① volume fraction of ethanol solution:49.96%; ② time of ultrosonic extraction:30 min and ③ ratio of mass of wormwood sample to volume of the ethanol solution:0.79 g to 15 mL. Rate of extraction found under this condition was found to be 1.09%. To verify the above optimized extraction condition, 3 portions (0.790 0 g each) of the powdered wormwood sample were taken and extracted under the optimized condition. The ethanolic mixture of solution and some undissolved was filtered and centrifuged. 1 mL of the supernatant was taken from each of the extract of the samples (the exact volume of each of the aliquot was determined from the estimated amount of flavonoids present in the sample solutions to have the mass concentration of the flavonoids in the aliquot happened to drop in the linearity range of the standard curve between 0.006 to 0.018 g·L-1) and proceeded as follows:2.38 mL of 5.3 g·L-1 Al(NO3)3 solution and 0.90 mL of pH 5.6 acetate buffer solution were added separately and the total volumes of the solutions were made up to 10.0 mL. After staying for completion of the fluorescent complexation between the flavonoids and Al(Ⅲ) ion values of intensity of the emission fluorescence were measured. It was shown that the average rate of extraction found was 1.08%, which gave a relative error of -0.92% with the theretically predicted value.
