Quality Evaluation for the Bark of Toricellia Angulata Oliv. Var. Intermedia (Harms) Hu in Different Collecting Seasons Based on HPLC Fingerprint and Chemical Pattern Recognition

    • The fingerprint for the bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu in different collecting seasons was established by HPLC. Combined with chemical pattern recognition method, the quality of the bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu in different collecting seasons was evaluated. After drying and sieving the fresh bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu in different collecting seasons, and active ingredients in the sample were extracted with methanol, and fingerprints of 11 batches of the bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu in different collecting seasons were established by HPLC. Chemometric softwares of "Similarity Evaluation System for Chromatographic Fingerprint of TCM" published by GPC (Version 2004 A), IBM SPSS Statistics 25 and SIMCA-P+12.0 were used for similarity analysis, hierarchical cluser analysis, PCA, and OPLS-DA. The results showed that 18 common peaks in the HPLC fingerprints of 11 batches of the bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu were found. RSDs of the relative peak areas between these common peaks and syringin peak were in the range of 30%-124%, indicating that the contents of chemical components in different collecting seasons were quite different. And the similarity degrees between fingerprints of 11 batches of samples and control fingerprint from chomatographic data of S6 were in the range of 0.932-0.982, indicating a relatively stable quality for the bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu in different collecting seasons. Meanwhile, 11 batches of the bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu could be divided into 2 categories by hierarchical cluster analysis, verified by results of PCA and OPLS-DA. Minor difference was found among the 11 baches of the bark of Toricellia Angulata Oliv. var. intermedia (Harms) Hu, owing to VIP values of the 18 common peaks all above 0.8, and the 7 substances corresponding to common peaks with VIP values greater than 1.0 were chosen as quality differential markers, to reflect of the changes of the main chemical composition of the bark of Toricellia Angulata Oliv.var.intermedia (Harms) Hu in different collecting seasons.
