Determination of Benzopyrene in Black Ginseng by Ultrasonic Extraction-Reversed Phase High Performance Liquid Chromatography with Fluorescence Detector

Abstract
The sample of black ginseng (0.500 0 g) was extracted ultrasonically for 0.5 h at 40℃ with 5 mL of n-hexane. The extract was purified by molecularly imprinted solid phase extraction column, and then determined by reversed phase HPLC with fluorescence detector, using Agilent TC-C18 column as stationary phase and 80% (φ) acetonitrile solution as mobile phase for elution. Under the optimized test conditions, linearity relationship between values of peak area and mass concentration of benzopyrene was found in the range of 0.50-20.0 μg·L-1, with detection limit (3S/N) of 0.20 μg·kg-1. Test for recovery was made by standard addition method, giving values of recovery in the range of 92.8%-97.5% and RSDs (n=6) of the determined values all less than 2.5%.
