Ion-chromatographic Determination of Saccharides and Polyols in Tobacco with Separation by Matrix Solid Phase Dispersion

    • Saccharides and polyols in tobacco sample were separated by the method of matrix solid phase dispersion and determined by ion chromatography (IC).The sample was mixed with graphitized carbon black granules in the mass ratio of 1 to 4 and ground in a motar to make it a homogeneous dispersion. The dispersion was then filled into a solid phase extraction column. The SPE column was eluted with hot water to dissolve the saccharides and polyols from the sample in the dispersion.CarboPac MA1 (4 mm×250 mm,5 μm) anion exchange column was used as stationary phase, and 0.5 mol·L-1 sodium hydroxide solution was used as mobile phase.Conductivity detection was adopted.Nine components in tobacco, including glycero1,propylene glyco1,xylito1,sorbito1,fructose,glucose,sucrose,maltose and galactito1 were completely separated within 50 min.Values of detection limits (3S/N) found were all below 50 μg·L-1.Tests for recovery and precision were made, giving values of recovery ranged from 94.6%-105.3%,and of RSD′s (n=7) less than 3.0%.
