    HPLC Determination of Catechin and Betulin in White Birch Bark

    • HPLC with UV detection at 205 nm was applied to determination of catechin and betulin in white birch bark.Shim-pack CLC-ODS C18 column (150 mm×6.0 mm,5 μm) was used as chromatographic column,and methanol-H2O mixture (75+25) was used as mobile phase,with flow-rate of 1.0 mL·min-1.Column temperature and column pressure taken were 30 ℃ and 7.747 MPa respectively.Linear relationships between values of peak area and concentration was found in the range of 20.0-400.0 mg·L-1 for catechin and in the range of 15.0-300.0 mg·L-1 for betulin.Precision was tested by analyzing a sample for 5 determinations,giving values of RSD′s 1.67% for catechin and 2.86% for betulin.Recovery was tested by standard addition method,values of average recovery found were 95.8% for catechin and 94.9% for betulin.
