    Separation of Oligosaccharides in Juice of Jerusalem Artichoke by Thin Layer Chromatography

    • Thin layer chromatography (TLC) was applied to the separation of oligosaccharides in juice of Jerusalem Artichoke.The silica gel plate (GF 254) was used as the stationary phase,and a mixed solvent,containing 1-butanol,2-propanol,acetic acid and H2O in the ratio of 7∶5∶2∶4,was used as the developing reagent.The colouring reagent was a mixed solution containing aniline (4 mL),diphenylamine (4 g) and conc.H3PO4 (20 mL) dissolved in 200 mL of acetone.Satisfactory separation of various kinds of oligosaccharides in juice of Jerusalem Artichoke was attained by a double development with the mixed solvent.The method proposed was feasible to be used for rapid TLC separation.
