Simultaneous HPLC Determination of Resveratrol,Polydatin and Free Emodin in Polygonum Cuspidatum

Abstract
HPLC with gradient elution,was applied to the simultaneous determination of resveratrol,polydation and free emodin in polygonum cuspidatum. Waters symmetry C18 column (3.9 mm×150 mm,5 μm) was used as chromatographic column. Mobile phase used was a mixed solution containing methanol and dil. phosphoric acid cH3PO4: 0.015 mol·L-1 and a flow-rate of 0.5 mL·min-1 was used in the elution. UV-detection at 284 nm was used in the determination. Pre-treatment of sample was performed by ultrasonic extraction with ethanol-H2O (1+1) and followed by 2 successive extraction with ethylacetate. Recovery was tested for resveratrol,polydatin and free emodin in polygonum cuspidatum grown in Jiangxi provicince,and the results obtained were 97.9%,96.1% and 95.6% respectively,with RSD′s less than 5%.
