    Resonance Scattering Spectrometric Determination of DNA with Polyamine as Probe

    • The interaction of DNA with three polyamines was studied by resonance scattering spectrometry. It was found that the intensity of resonance scattering of spermine was greatly enhanced by calf thymus DNA in a Britton-Robinson buffer of pH range of 2.21-9.15. A resonance scattering peak was found at 343 nm. The concentration of calf thymus DNA in the range of 0.1-40 mg·L-1 was proportional to the enhanced resonance scattering intensity,with a correlation coefficient of 0.998 9. The detection limit was found to be 19 μg·L-1. The proteins at 10 mg·L-1,nucleotides at 10 mg·L-1 did not interfere with the determination of 2.0 mg·L-1 DNAs. Most of the common metal ions at 1.0×10-6mol·L-1 were tolerable. Based on these facts a method for determination of DNA by resonance scattering spectrometry was proposed.
