    CE Determination of Trigonelline in Rabbit Serum

    • CE was applied to the determination of trigonelline in rabbit serum, and pharmacokinetics of trigonelline within rabbit body was studied. Analytical conditions were studied and following optimized parameters were given: ① electrophoretic medium: citrate buffer solution of pH 5.6; ② wavelength for detection: 224 nm; ③ separation voltage: 22 kV; ④ time of sample introduction: 10 s; ⑤ temperature of capillary: 25 ℃. Linear relationship between values of peak height and mass concentration of trigonelline was kept in the range of 0.1-100 mg·L-1, with detection limit (3S/N) of 0.05 mg·L-1. The proposed method was used in the analysis of sample of rabbit serum, by using the PKS program to fit the plot of drug concentration in blood vs. time of drug given to obtain a series of pharmacokinetic parameters. It was found that the metabolic process of trigonelline in rabbit blood was conformed with the characteristic of two-compartment open model, and that the drug was absorbed at a middle rate and eliminated at a rapid rate.
