    Determination of Saccharides in Food by Ion Exchange Chromatography with Integral Pulse Amperometric Detection

    • Ion exchange chromatography with integral pulse amperometric detection was applied to the determination of glucose,fructose,sucrose,lactose and maltose in food.Retention property of the 5 saccharides in the elution with NaOH solution was studied.CarboPacTM PA 10 column was used as separation column,and mixtures of pure water and 0.2 mol·L-1 NaOH solution in different ratio were used as mobile phase in the gradient elution.The flow rate taken was 0.5 mL·min-1.In the amperometric detection,gold electrode was used as working electrode,Ag/AgCl electrode was used as reference electrode.Under the optimum conditions,the 5 saccharides in a sample were separated and determined effectively and simultaneously.Linearity ranges for all the 5 saccharides were obtained in the range of 0.5-10.0 mg·L-1.Detection limit (3S/N) found were in the range of 0.8-2.5 μg·L-1.Test for recovery was made by addition of the standards of the 5 saccharides to a substantial sample as the matrix,values of recovery obtained were in the range of 91.1%-104.8%.
