    RP-HPLC Determination of Panaxadiol in Ginseng with Pre-column Derivatization

    • A new method of RP-HPLC with precolumn derivatization was proposed for the determination of panaxadiol in ginseng.Benzoyl chloride was used as derivatizing agent.The Hypersil ODS2 column (4.60 mm×150 mm,5 μm) was used as stationary phase,and a mixed solvent of acetonitrile and water (40+60) was used as mobile phase.UV-detection at the wavelength of 230 nm and the external standard method were adopted in the determination.Linear relationship between values of peak area and mass concentration of panaxadiol (in form of its derivatized product) was obtained in the range of 0.01-0.10 g·L-1.Detection limit (3S/N) found for this method was 0.07 mg·L-1.Precision of the method was tested with 0.06 g·L-1 of the standard panaxadiol solution for 6 replicate determination including the precolumn derivatization process,value of RSD calculated from data of peak are was 1.61%.
