    HPLC Determination of Diethylstibestrol in Chicken

    • High performance liquid chromatography (HPLC) was applied to the determination of residual diethylstibestrol in chicken as food. Smashed and homogenized chicken sample was extracted ultrasonically with methanol for several times as directed in the procedure. The extracts were combined and evaporated to near dryness. The residue was taken up with 1 mL of methanol and the solution was used for HPLC determination. Symmetry C18 was used as the chromatographic column and a mixture of methanol and 0.012 8 mol·L-1 NaH2PO4 solution mixed in the ratio of 63 to 37 ( by volume),adjusted to pH 5,was used as the mobile phase at a flow-rate of 1.0 mL·min-1. UV-detection at 230 nm with array photodiode was used in the determination. Detection limit (S/N=3) was found to be 20 μg·kg-1. Sample of chicken was analyzed by this method and values of RSD′s (n=5) and recovery were found to be less than 10% and in the range of 87.5% to 92.3% respectively.
