    Fluorospectrophotometric Determination of 1-Hydroxypyrene in Human Urine with Micellar Sensitization of Triton X-100

    • It was found that in a B-R buffer medium of pH 8.0,the fluorescence intensity of 1-hydroxypyrene (1-OHP in short) was significantly sensitized by the Triton X-100 micelle,and linear relationship was obtained between values of fluorescence intensity and mass concentration of 1-OHP in the range from 3.2 to 982.0 μg·L-1 when measured at the wavelengths of (λex) 279 nm and (λem) 390 nm.Detection limit (3S/N) of the method found was 1.0 μg·L-1.Based on these facts,a method for determination of 1-OHP in human urine was proposed.Precision of the method was tested by replicate determination,and values of RSD (n=7) found were 1.78% for 200 μg·L-1 of 1-OHP and 0.98% for 600 μg·L-1 of 1-OHP.Recovery was tested by standard addition method on the base of human urine samples as matrixes,and values of recovery obtained were ranged from 93.2% to 100.7%.Results obtained by the present method were checked very well with the results found by HPLC.
