GC-MS Determination of Fatty Acids in Tissues of Hynobius leechii

Abstract
Crude fatty oil of various tissues (skin, muscle and viscera) of freeze-dried Hynobius leechii were extracted separately with petroleum ether in Soxhlet′s extractor. The extracts were washed separately with saturated NaCl solution and distilled water, and dehydrated overnight with anhydrous Na2SO4. The solvent in the extract was removed by rotary evaporation, and 3 portions of oil separated from the tissues of Hynobius leechii were obtained. An aliquot of 0.20 mL was taken and esterified with methyl alcohol in KOH solution and extracted with 1.0 mL of mixture of ethyl ether and n-hexane (2+1), 0.4 mL of the supernatant was taken and analyzed by GC-MS. In portion of oil from the muscle, 21 fatty acids were identified, in which oleic acid and linoleic acid were the main components, and the total of unsaturated fatty acids amounted to 66.26%. In the oil from the skin, 8 fatty acids were identified with oleic acid and linolenic acid as the main components, and with total amount of unsaturated fatty acids achieved to 68.79%. In the oil from viscera, 9 fatty acids were identified with oleic acid, linoleic acid and palmitic acid as main components, and with the total of unsaturated fatty acids amounted to 65.64%.
