HPLC Determination of Puerarin in An Health-care food “Kudzu Vine Root Powder”

Abstract
Concents of puerarin in kudzu vine root powder produced in different areas in our country were determined by HPLC.The Agilent-C18 column (4.6 mm×300 mm,5 μm) was used as the stationary phase,and a mixfure of methanol and H2O,mixed in the volume ratio of 70 to 30,was used as the mobile phase with a flow-rate of 1.0 mL·min-1.Volume taken for sample introduction was 10 μL.UV-detections were made at the wavelength of 250 nm.Linear relationship was obtained in the range of 10.00 to 100.00 mg·L-1 between the values of peak area and concentration of puerarin,with its detection limit (3S/N) of 1.0 mg·L-1.Test for precision was made at 3 different concentration levels of 1.58,1.55 and 0.24 mg·L-1 for 6 determinations each,giving values of RSD′s of 0.57%,0.64% and 1.71% respectively.Under the condition of sample analysis,recovery of the method was tested by addition of standard solution of puerarin at 3 different concentrations,and values of recovery (n=6) obtained were in the range of 99.0% to 99.8%
